Structure and mechanism of action of botulinum and tetanus neurotoxins (literature review)

چکیده

Botulinum neurotoxins and tetanus are the strongest known toxins that cause neuroparalytic syndromes in botulism tetanus. This review aimed to systematize scientific data on structures mechanism of actions botulinum neurotoxins. proteins containing functional domains responsible for receptor binding, transmembrane translocation, proteolytic cleavage required exocytosis synaptic vesicles release neurotransmitters into cleft. The main stages neurotoxin action include binding presynaptic membrane, internalization bound toxin cytosol via endocytosis, translocation L-chain HN domain, disruption interchain disulfide bond with express its catalytic activity (as a metalloprotease) cytosol, selective one or more soluble N-ethylmaleimide-sensitive factor attachment complex subsequent blockade neurotransmitter release.

The mechanism of action of tetanus and botulinum neurotoxins.

Tetanus and botulinum neurotoxins, produced by anaerobic bacteria of the genus Clostridium, are the most toxic proteins known and are the sole responsible for the pathogenesis of tetanus and botulism. They enter peripheral cholinergic nerve terminals and cleave proteins of the neuroexocytosis apparatus causing a persistent, but reversible, inhibition of neurotransmitter release. Botulinum neuro...

Central effects of tetanus and botulinum neurotoxins.

Tetanus neurotoxin (TeNT) and botulinum neurotoxins (BoNTs; from A to G) are metalloproteases that act on nerve terminals to prevent exocytosis. They are extensively exploited for the study of cellular physiology. Moreover, BoNTs are also employed in clinical neurology for the treatment of several disorders characterized by hyperexcitability of peripheral nerve terminals. This review summarizes...

Aluminum Phosphide Poisoning, Mechanism of Action and Treatment: a Literature Review

Aluminum phosphide (AlP) is an important fumigant, a commendable and very effective outdoor and indoor insecticide. AlP, locally named “rice tablet”, is widely used to protect rice. As soon as taking a very small amount of an AlP tablet, phosphine vaporizes due to the exposure to the air and affects different kinds of organs. Although, in most cases, clinical history can help making the final d...

Mechanism of action of neurotoxins.

This paper is a summary of studies over the past few years that pertain to animal neurotoxins. These toxins are found throughout the animal kingdom. Homologies exist in the structures of these poisons within classes and point to conservation of active sites throughout evolution. In the case of the peptides, invariant amino acids may be involved in the active site, be essential for maintaining t...
